It is characterized by the accumulation of an abnormal isoform (PrP Sc ) of host encoded prion protein (PrP C ) in the central nervous system which leads to progressive neurodegeneration and death. Scrapie represents the prototype of the so-called prion diseases. It is observed to date as two types, classical and atypical scrapie. The susceptibility to both types is modulated by polymorphisms of the prion gene. Whereas classical scrapie is clearly a naturally occurring transmissible disease, atypical scrapie may also be caused by the spontaneous misfolding of prion protein. This review gives an overview on the current knowledge of classical and atypical scrapie in sheep and goats with special emphasis on epidemiology, clinical and pathological signs, genetic susceptibilities, diagnosis and the characteristics of the most common scrapie strains.

Diversity in Neuroanatomical Distribution of Abnormal Prion Protein in Atypical Scrapie

Scrapie is a transmissible spongiform encephalopathy (TSE) in sheep and goats. In recent years, atypical scrapie cases were identified that differed from classical scrapie in the molecular characteristics of the disease-associated pathological prion protein (PrP(sc)). In this study, we analyze the molecular and neuropathological phenotype of nine Swiss TSE cases in sheep and goats. One sheep wa...

Clinical Examination Protocol to Detect Atypical and Classical Scrapie in Sheep

The diagnosis of scrapie, a transmissible spongiform encephalopathy (TSEs) of sheep and goats, is currently based on the detection of disease-associated prion protein by post mortem tests. Unless a random sample of the sheep or goat population is actively monitored for scrapie, identification of scrapie cases relies on the reporting of clinical suspects, which is dependent on the individual's f...

The placenta shed from goats with classical scrapie is infectious to goat kids and lambs

The placenta of domestic sheep plays a key role in horizontal transmission of classical scrapie. Domestic goats are frequently raised with sheep and are susceptible to classical scrapie, yet potential routes of transmission from goats to sheep are not fully defined. Sparse accumulation of disease-associated prion protein in cotyledons casts doubt about the role of the goat's placenta. Thus, rel...

Distinct proteinase K-resistant prion protein fragment in goats with no signs of disease in a classical scrapie outbreak.

Considerable efforts have been directed toward the identification of small-ruminant prion diseases, i.e., classical and atypical scrapie as well as bovine spongiform encephalopathy (BSE). Here we report the in-depth molecular analysis of the proteinase K-resistant prion protein core fragment (PrP(res)) in a highly scrapie-affected goat flock in Greece. The PrP(res) profile by Western immunoblot...

Sparse PrPSc accumulation in the placentas of goats with naturally acquired scrapie

BACKGROUND Domestic goats (Capra hircus) are a natural and experimental host of scrapie and bovine spongiform encephalopathy, the transmissible spongiform encephalopathies (TSE) of sheep and cattle. Goats are also susceptible to experimental infection with the agents of TSEs of deer and elk (chronic wasting disease) and humans (Creutzfeldt Jakob disease). Distribution of PrPSc, the abnormal pri...
